How border security 'trigger' could stop immigration reform

Congressional negotiators say immigration reform will need a border security 'trigger' to pass. But agreeing on what counts as 'border security' won't be easy, and could determine whether reform happens.

What?s Congress to do?

Figure out a ?trigger,? where advances in border security are deemed sufficient to trigger the beginning of the journey to citizenship for the undocumented already in the country.

As immigration reform negotiations continue, determining just what counts as a ?secure border? and how to link that to plans for the undocumented will be crucial. Indeed, finding an answer could determine whether a bipartisan immigration reform measure reaches President Obama?s desk or if 2013 is yet another disappointment for reformers.

Historically, those on Capitol Hill have tried to craft a delicate balance between border security and a path to legal status for the undocumented. For example, the comprehensive immigration reform legislation of the George W. Bush years, which ultimately failed, had a series of triggers. In 2009, Sen. Chuck Schumer (D) of New York proposed more broadly that ?operational control? of the border ?must be achieved within a year of enactment of legislation.?

But those triggers aren't helpful anymore. Most of the benchmarks for border security established in 2007, for example, have been met today, according to an analysis?by the pro-reform advocacy group America?s Voice.

Border patrol staffing north of 20,000? Check: there are more than 21,000 agents on the border at present. Requirements for unmanned drones and a variety of other observation methods? All are at or above the 2007 requirements today. Fencing? Within eight miles of the 2007 target.

Department of Homeland Security Secretary Janet Napolitano said on Tuesday that while the department lacks a single measure on which to base a trigger ? and that a trigger based on any single measure would be a bad idea ? all the data DHS collects point to a border safer than ever before.

Some Democrats and immigration reform advocates take this to say that the border is secure already and should not stand in the way of the undocumented becoming US citizens even if further border security measures are needed.

Republican reformers like Sen. John McCain of Arizona have a slightly different view, holding that while the southern border certainly is far improved from nearly a decade ago there?s still plenty of room for improvement.

?There's no question there's been a significant reduction in illegal crossings over the past five years?. But that work is not yet complete,? Senator McCain said in January at the press conference announcing a bipartisan Gang of Eight?s principles.

Other Republicans, like Sen. Jeff Sessions (R) of Alabama, are skeptical of such claims and believe figuring out just what constitutes a secure border should be among the goals of a lengthy series of hearings around immigration reform.

So what do lawmakers propose to do this time around? Mr. Obama?s answer appears to be scrap the trigger altogether.

His immigration statements have notably left out any linkage between border security and permanent legal status for the undocumented, noting in his immigration reform plan that if an undocumented person meets certain criteria including paying fines, learning English, and waiting until all other current prospective immigrants have passed through the immigration system, ?there will be no uncertainty about their ability to become US citizens.?

However, White House spokesman Jay Carney told reporters something far short of ?no triggers? when asked directly about the subject on Tuesday: ?There are a variety of metrics by which you can measure, and we do measure, progress on border security.? We're working with Congress on this, with the Senate on this. Progress has been made.?

In the Senate, the Gang of Eight requires that a future group of Southwest border leaders, including governors, attorneys general, and community figures, would certify that the border is secure before any undocumented immigrants could receive green cards. Essentially, the undocumented would, after meeting eligibility steps similar to the president?s, be allowed to receive legal status in the country. Then, when the group of border leaders certified the border secure, those immigrants could begin the path to citizenship.

The reform outline of Sen. Rand Paul (R) of Kentucky calls for a Capitol Hill-based verification scheme that would require Congress to certify that the border is secure for five consecutive years but would begin offering temporary work visas to the undocumented in the second year after enactment.

First quarter report cards: Which fighters rose above early in 2013?

Sunday will mark the end of the first quarter. It's been a busy three months in MMA, so it's a good time to look back at the past three months and see who rose above the rest.

Biggest upset: Did you expect to see Robbie Lawler walk away with the win over Josh Koscheck at UFC 157? What about Antonio Rogerio Nogueira beating former UFC light heavyweight champ Rashad Evans? But there's no question who scored the biggest upset of the first quarter of 2013. Antonio Silva's TKO of Alistair Overeem at UFC 156 left more than Overeem's jaw on the floor.

Best knockout: Vitor Belfort's knockout of Michael Bisping at the January UFC on FX was memorable, as was Tyron Woodley's 36-second dismantling of Jay Hieron at UFC 156. Wanderlei Silva, Robbie Lawler and a slew of Bellator fighters have posted impressive knockouts. But the one that had everyone talking was on "The Ultimate Fighter." Uriah Hall's knockout of Adam Cella* was perfectly executed, and had the rest of the TUF house worried about Hall's skills. For that, Hall wins knockout of the first quarter.

* The knockout actually occurred last year, but aired in the first quarter of 2013.

Best submission: For the best tapouts of the first three months of 2013, Bellator provided a nominee when Dave Vitkay choked out Jesse Peterson in just 18 seconds. Gabriel Gonzaga's guillotine of Ben Rothwell stands out, as is Ronda Rousey's arm bar of Liz Carmouche at UFC 157. The winner comes from the undercard of UFC 157 as Kenny Robertson managed a weird yet incredibly painful looking kneebar-esque submission of Brock Jardine. It was the kind of submission that left MMA fans wondering what had happened.

Best rising star: There were plenty of candidates for fighters who really made an impressive leap in the MMA world early in 2013. Jordan Mein's UFC debut at UFC 158 included rolling out of an arm bar with Dan Miller, and then knocking him out before the first round ended. Abel Trujillo's TKO win by knees to the body of Marcus Levesseur was pretty fantastic, too. But it's Trujillo's opponent at UFC 160, Khabib Nurmagomedov who is taking home the prize. His TKO of Thiago Tavares in Brazil showed he can fight well under any conditions.

Best fight: Johny Hendricks and Carlos Condit put on a great scrap at UFC 158. Brian Stann and Wanderlei Silva's bout was thrilling before Stann was knocked out. Demetrious Johnson and John Dodson's title fight at UFC on Fox 6 was memorable, as was Frankie Edgar and Jose Aldo's title bout at UFC 156. The one fight that stood out above the rest, though, was Dennis Bermudez and Matt Grice's fight from UFC 156. Throughout the bout, both fighters should have given up. Both fighters should have been knocked out. But neither man gave in. Bermudez won the decision, but both fighters will be remembered for this bout.

"The Host" review: Invasion of the boring snoozers

LOS ANGELES (TheWrap.com) - Here we go again: No sooner did the "Twilight" franchise pack up and head to that shiny coffin in the sky that we get another Stephenie Meyer story about the Special-est Girl in the World and the cute boys who fall madly in love with her. No bloodsuckers or lycanthropes this time, however; now it's all about glittery dust-bunny aliens who have taken over all the human bodies and turned them?polite?

"The Host" posits an alien invasion whereby almost all of humanity has been body-snatched, resulting in a planet that's clean, well-fed and kind. A remaining group of people has decided not to swap their souls for this utopia; one of them is Melanie (Saoirse Ronan), who's been on the run with her younger brother Jamie (Chandler Canterbury) and her boyfriend Jared (Max Irons), whom they encountered along the way.

To keep the Seeker (Diane Kruger) and her troops away from Jamie, Melanie distracts them and jumps out of a window. The aliens restore her body (through some of sort of spray-based technology) and implant it with a thousand-year-old "soul" known as The Wanderer, but Melanie's sheer force of will remains alive, so the two of them duke it out over control of the body, like a not-remotely-funny version of Steve Martin and Lily Tomlin in "All of Me."

Melanie convinces Wanderer to escape and to go to the hidden underground colony run by Melanie's uncle Jeb (William Hurt); Jamie and Jared are there, as are brothers Ian (Jake Abel) and Kyle (Boyd Holbrook). While Jeb slowly intuits that Melanie's spirit still lives inside her alien-possessed body, Ian starts falling for Wanderer (or "Wanda," as Jeb calls her) while Jared refuses to believe that his beloved might still be sharing space with the visitor from another world.

During the dull stretches of "The Host" - and, sadly, there are many of them - you may find yourself rewriting the movie as a French farce, with two men in love with the two personalities inside the same woman (while a third man, Kyle, wants to kill her). Ideally, your version wouldn't cast three male leads who couldn't be distinguished in a police lineup; the marked resemblance between Jared, Ian and Kyle makes for occasional confusion and an extra helping of blandness that this flat movie certainly didn't need. (Say what you will about the thespian talents of Robert Pattinson and Taylor Lautner, at least you could tell them apart.)

A more competent movie could slip in some kind of subtext about young women being at war with their own bodies and desires (when Wanda kisses a boy, Melanie gets mad and slaps him), but writer-director Andrew Niccol doesn't plumb any of that from Meyer's book.

Niccol's expertise is films about future dystopias with very attractive flies in the ointment ("Gattaca," "In Time"), but he fails to bring any excitement here, from either plot or characters or the action, the latter of which always seems to involved chrome-covered cars, helicopters and guns.

There are so many moments of unintentional humor in the film, that the intentional jokes fall a bit flat, because you're not sure if they're supposed to be funny or not.

If you saw "Atonement" or "Hanna," you know that Ronan is a fine young actress, but most of the role calls for her to argue with her own voiceovers, and it gets increasingly hard to watch. Perhaps having a wraith version of Melanie squabbling with the tangible Wanda might have played better. The guys in her orbit have performances that are as interchangeable as their blankly handsome faces.

None of the humans pops as an interesting character, so it's hard to get upset about the alien invasion when everyone on screen seems to be on the same uninteresting wavelength. There's not even a marked enough difference between Melanie and Wanda-in-Melanie's-body to make us care if our heroine gets her own body back.

There's some nifty art direction here, notably the labyrinth of caves where the humans are hiding out, but "The Host" feels so polished and passionless that one might think that it was the product of one of the bland aliens. And if that's the case, all the more reason to banish them from the planet.

How To Market Your Business For Free Using Twitter | Content for ...

Today we're going to discuss how you can build your business for free using Twitter. Twitter has grown to be one of the largest social media networks on the internet, and is actively visited by celebrities and high-profile people.

If you want to put Twitter to work for your business, you first need to create an account on Twitter. Register for free and upload a picture and a short description of yourself or your business. Make sure you include your main website in your bio.

Next thing you want to do is to write and post around 20 tweets. This is so your account doesn't look empty when people visit it to check if you are "follow-worthy".

All right, now that you got an account you need to get yourself a good Twitter client so that you don't have to use the normal Twitter interface (which kinda sucks.)

I strongly recommend that you Google "HootSuite", which is a free website that manages your Twitter account.

Spend some time getting yourself familiar with HootSuits interface, and then setup your "columns" over there like this:

Column #1: Sent tweets

This is your tweet stream. Keep this one handy to know what you just said.

Column #2: Mentions

It will track who is mentioning you and replying to your tweets. It's important to respond to these occasionally and to respond to them quickly. This will ensure that the people you interact with visit your profile and start following you.

Column #3: Influencers list

It's important to know who your influencers are and what they are tweeting, so you can get on their radar. You could follow your influencers, but I don't like to until there's been some interaction.

Instead, I use private lists, the Twitter-feature to keep tabs.

Here's how to set them up in HootSuite:

Step 1: Click on "Add Stream."
Step 2: From the pop-up menu, select "Lists."
Step 3: Select your profile from the drop-down menu.
Step 4: Select the "Create a New List" radio button.
Step 5: Name the list "Influencers."
Step 6: Add a description (optional).
Step 7: Click the "Add Stream" button.
Step 8: Once the stream is added, click the triangle on the top of the column.
Step 9: From the drop-down, select "Preferences."
Step 10: Add users to your list by typing in their usernames and clicking "Add."

Make sure to set this list as "private", otherwise everyone can see it, and it will notify who ever's on the list.

Column #4: Scheduled tweets

I use this column to keep tabs on what I've scheduled ahead of time to know what's coming and if I need to add more tweets or perhaps adjust my Tweet Schedule.

Column #5: My tweets, retweeted

The My Tweets, ReTweeted column shows you who is retweeting your tweets.

Column #6: Search filters

The best thing about HootSuite is that you can monitor search results in real-time. We're going to save those in the next set of columns.

My next step, while interacting with Twitter influencers is courting potential followers. It's also stupid simple. All I did was monitor the real-time keyword and hashtag searches I setup earlier, and reply to people's tweets. Most of the time it was answering questions, but I found an insightful comment or even an encouraging message beneficial.

As soon as you see something interesting in the real-time search column, be sure to answer it. Do this every day for 30-60 minutes and you will gather a large following of twitter fans in no time.

After that you can just post links to your website in between all the cool "normal" things that you tweet to your followers. Also, make sure that you track the traffic coming to your site from Twitter, so that you know how many visitors you are getting from your efforts. Try this and you will start to see visitors coming to your website from Twitter every time you post a tweet that includes a link back to your website.

The New York International Auto Show is in full swing at the Jacob Javits Center on Manhattan's West Side, and journalists have been treated to the debuts of more than a few remarkable vehicles.

But while most of the automakers have hired female models ? AKA booth babes ? to stand alongside their products, Bugatti took a different approach this year.?

Accompanying the Veyron, the world's fastest production car, is a security guard, whose job consists of keeping interested members of the press a few yards away from the multi-million dollar ride.

According to the guard, someone checking out the Veyron at last year's show accidentally damaged it, costing the company quite a bit of cash. So for 2013 they're not taking any chances ? although rumor has it journalists at the show can get closer if they talk to the Bugatti media representative, and set up a time beforehand.

Democratic leader says assault weapon ban lacks support in Senate

WASHINGTON (Reuters) - Senate Majority Leader Harry Reid, a Nevada Democrat, said on Tuesday that fewer than 40 of his chamber's 100 members support a White House-backed bill to renew a ban against military-style assault weapons.

The scant support virtually assures rejection of the bill, a centerpiece of Obama's bid to curb gun violence in wake of a massacre at a Connecticut elementary school on December 14 that left 20 children and six adults dead.

"Right now," Reid told reporters, the bill drafted by Democratic Senator Dianne Feinstein of California, "has less than 40 votes."

A simple majority would be needed to win passage of the bill in the Democratic-led Senate, but 60 would likely be required to first clear an anticipated Republican procedural roadblock.

Obama, Feinstein and others backers of the bill to renew a ban against the sale and manufacture of military-style assault weapons, argue that the firearms should be removed from the street.

But opponents, a number of Senate Republicans, along with some Democrats from rural areas where guns are popular, argue that the action would amount to a violation of the right to bear arms, which is enshrined in the U.S. Constitution.

Democrats control the chamber, 55-45, meaning that a number of Reid's fellow Democrats have made it clear that they intend to oppose the bill.

Americans sympathize with Israel, want U.S. out of it

Many more Americans continue to side with Israel rather than the Palestinian Authority, but - with President Obama's first visit there days away - most also prefer to leave peace negotiations to the two protagonists, rather than having the United States take the lead.

Fifty-five percent in this ABC News/Washington Post poll sympathize more with Israel, vs. 9 percent who side more with the Palestinian Authority, with the rest favoring neither, or undecided. It's been a similar gap for many years, including polling back to the 1980s testing Israel vs. the Arab nations of the Middle East.

Despite that preference for Israel, seven in 10 want the U.S. largely to leave resolving the conflict to the Israelis and Palestinians themselves - a result that underscores the difficulties in finding a solution to the decades-old conflict. Preference for the United States to eschew a leading role is 15 percentage points higher than the last time it was asked in an ABC/Post poll, during an outbreak of violence between the two sides nearly 11 years ago.

Even among those who are more sympathetic to one side or the other, regardless of which side it is, about two-thirds don't want the U.S. to take the leading role. That preference rises to about three-quarters of those who don't favor either side.

In another expression of support for Israel, more Americans say the Obama administration has put too little pressure on the Palestinian Authority than too much pressure - 34 vs. 8 percent. They split about evenly, by contrast, on whether the administration has put too much or too little pressure on Israel. About four in 10, meanwhile, think the U.S. has appropriately pressured each side in the conflict.

RELIGION and POLITICS - This poll, produced for ABC by Langer Research Associates, finds little sympathy for the Palestinian Authority across groups, always well behind support for Israel. However, breadth of support for Israel varies considerably, with the Palestinian Authority and frustration with both groups gaining somewhat among less broadly pro-Israel groups.

Among religious groups, sympathy for Israel peaks, at 76 percent, among evangelical white Protestants, falling to 55 percent among non-evangelical white Protestants and Catholics, and bottoming out at 39 percent among those who aren't religiously affiliated. Religiosity is a factor as well, with those who attend religious services more apt to side with Israel.

Support for Israel also is broad - more than seven in 10 - among Republicans and conservatives alike. This drops to roughly five in 10 moderates, independents and Democrats, and to just 39 percent of liberals, with more saying they favor neither side, compared with Republicans and conservatives.

A similar pattern plays out on the matter of the Obama administration's use of influence on each side, with Republicans and conservatives more likely than others to think Israel is being pressured too much and the Palestinian Authority too little. Majorities of Democrats, not surprisingly, are happy with the pressure the Obama administration's applying to each side.

On the other hand, when it comes to U.S. involvement in the peace process, there's agreement across religious, partisan and ideological groups (from 66 to 70 percent) that the two sides should handle negotiations themselves.

Age is another prominent marker of support for Israel, ranging from 48 percent among younger adults to 57 percent of 40- to 64-year-olds and topping out at two-thirds among seniors. Views that the Obama administration is putting too much pressure on Israel, and is applying too little of its muscle with the Palestinian Authority, also peak among seniors.

Obama is scheduled to leave Wednesday for a two-day visit to Israel and the West Bank, ruled by the Palestinian Authority.

METHODOLOGY - This ABC News/Washington Post poll was conducted by telephone March 7-10, 2013, among a random national sample of 1,001 adults, including landline and cell-phone-only respondents. Results have a margin of sampling error of 3.5 points, including design effect. Partisan divisions are 33-25-35 percent, Democrats-Republicans-independents.

Thankfully, my own feeble attempts to play Cut The Rope using the Leap Motion Controller weren't recorded on camera, but we did film a short demo by Vice President of Product Marketing Michael Zagorsek. He showed off a 3D visualizer that helps developers understand the controller's capabilities, then played Fruit Ninja using a chopstick, and finally used the controller to sculpt a digital clay.

Better Support Needed for Victims of Intimate Partner ViolenceA new study finds that when partner violence does occur, only about one in five women are asked or counseled by a health care provider about abuse.

The findings come from a study by Penn State University researchers that advocates the need for improved identification and counseling about intimate partner violence.

?Our research shows that we (as a health care community) haven?t been doing a good job of identifying and counseling about intimate partner violence,? said Dr. Jennifer S. McCall-Hosenfeld, a primary care physician and assistant professor of medicine and public health sciences.

?Only 20 percent who had been exposed to intimate partner violence received safety and violence counseling in the two years following the abuse, and only 11 percent of all women had discussed violence and safety at home with a health care provider.?

McCall-Hosenfeld and colleagues examined preventive healthcare services for women of reproductive age and how exposure to intimate partner violence (IPV) was associated with relevant preventive healthcare services.

The study focused on women who had reported experiencing IPV ? specifically physical violence, sexual violence and threats of either by a current or former partner or spouse.

More than a third of the women in the U.S. have experienced IPV, according to the National Intimate Partner and Sexual Violence Survey conducted by the Centers for Disease Control and Prevention.

The researchers point out that this abuse can lead to serious immediate and long-term health problems, as a result of inadequate preventive health care services.

Between 2004 and 2007 McCall-Hosenfeld and colleagues surveyed 1,420 women of reproductive age who were participating in the Central Pennsylvania Women?s Health Study.

The women responded to questions regarding intimate partner violence, healthcare access, socio-economic status and whether they had received preventive healthcare services.

Study results are published in the March/April issue of Women?s Health Issues.

In the first survey the researchers established whether women had recently been exposed to IPV by asking them to respond ?yes? or ?no? to a series of questions. Each question was preceded by, ?In the past 12 months, has a spouse, partner or boyfriend?? and completed with statements including: ?Threatened to hit you or throw something at you?? and ?Pushed, grabbed, shoved or slapped you??

Two years after the first survey the researchers followed up with the women, asking about any preventive health care services and preventive counseling they had received over the past 24 months.

Again, the women surveyed were asked to respond ?yes? or ?no? to a series of questions, including: ?Have you been tested for sexually transmitted infections or HIV?? and ?Has a doctor or health professional asked you or talked to you about concerns about safety or violence in your home??

According to the study the findings represent ?a missed opportunity for health promotion among women exposed to IPV.?

?Our data suggest that many women who have been exposed to IPV are not being appropriately identified in health care settings and are not getting many of the health care services they need,? said McCall-Hosenfeld.

In January 2013 the United States Preventive Services Task Force released a guideline recommending that clinicians screen all women of reproductive age for IPV and help provide intervention services when necessary.

?We have a long way to go for healthcare to be in compliance with this guideline,? said McCall-Hosenfeld.

?We?ll need to have a culture shift in many settings so that healthcare providers are comfortable with asking patients about IPV, patients are comfortable with being asked and the health care systems work to ensure that all patients get the services they need.?

Researchers say that although this study does not specifically address how to improve preventive services for those who have experienced IPV, the need is now documented with future research to continue along this thread.

TRIPOLI, March 11, 2013 (AFP)

At least 51 people have died in Libya since Saturday after drinking homemade alcohol, with another 378 suffering from
alcohol poisoning, the Libyan health ministry said on Monday.

It said 38 people died in Tripoli hospitals while 13 others perished on their way to neighbouring Tunisia, where their families were hoping to provide them with medical care.

At least 378 people were poisoned after drinking homemade alcohol containing methanol, the ministry said, urging Libyans to stop consuming alcoholic beverages.

Interior Ministry spokesman Houssine al-Ameri told AFP an investigation had been launched to determine who was responsible for the killer booze.

A security official, who declined to be named, added that the authorities were preparing to crackdown on suppliers and traffickers of alcohol, whose sale and consumption is banned in Libya although it can be found on the black market.

Tripoli medical centre?s health chief Youssef al-Wafi said on Sunday that ?preliminary tests revealed the symptoms of poisoning from the consumption of homemade alcohol containing methanol.?

Wafi said the health risks included kidney failure, blindness, seizures and possible death, with those thought to have consumed the toxic drink ranging in age from 19 to 50.

The consumption and sale of alcohol is banned in Libya, even though it is available on the black market.

With its porous borders, Libya has seen a significant increase in drug and alcohol trafficking since the now-slain dictator Moamer Kadhafi?s regime was ousted in 2011.

You own a car in Greenwich, you?re charged less than 11 mills. You own a car in Hartford, you?re charged 75 mills. It makes no sense. A car is a car is a car.?

Why is a 2,000 square foot house in Bridgeport taxed at a higher mill rate than one in Darien? A house is a house is a house, no? Shouldn?t the Guv and lawmakers be tackling that? After all, while property values are down in Bridgeport the taxes are up. So it makes it difficult to own a home even in our most affordable communities.

The Guv is on to something, there is a fairness issue related to taxes, but it?s an issue that?s been around now for decades and no one in Hartford has actually shown any backbone when it comes to doing something about it other than creating blue ribbon task forces to study the issue.

A town councilman in Trumbull called The Mines Tuesday to talk about the stock market, but was sucked into the car tax proposal, calling it a dumb idea as it?s just shoveling the burden from car owners to property owners.

?The only way to solve the tax problem (car and property) is to look at the entire taxing system as a whole,? said Michael London, a member of the Trumbull Town Council, said there?s no question

But it?s an important one. Bridgeport Mayor Bill Finch and others have rightly pointed out they need money for cops, firemen, parks and other services that people need. It would be interesting to see how much it costs to have a cop on a beat in Greenwich compared to other communities.

Where is the difference in costs for these communities? Theoretically, Bridgeport should be able to spread out the costs of its services among more people and offer a competitive tax rate to suburbs. But it can?t for some reason.

What the state does or doesn?t do matters in the towns and cities. And while Finch and other mayors? concerns are legitimate, the answer to providing services isn?t always just to stop doing it. If that were the case the libraries would have been closed years ago, but most of them have continued to function by leveraging volunteer resources and fund raising.

Ultimately, it?s time for the state and cities to get creative and to leverage their best resources, the people who live here.

Researchers with the U.S. Department of Energy (DOE)'s Lawrence Berkeley National Laboratory (Berkeley Lab) have achieved a major advance in understanding how genetic information is transcribed from DNA to RNA by providing the first step-by-step look at the biomolecular machinery that reads the human genome.

"We've provided a series of snapshots that shows how the genome is read one gene at a time," says biophysicist Eva Nogales who led this research. "For the genetic code to be transcribed into messenger RNA, the DNA double helix has to be opened and the strand of gene sequences has to be properly positioned so that RNA polymerase, the enzyme that catalyzes transcription, knows where the gene starts. The electron microscopy images we produced show how this is done."

Says Paula Flicker of the National Institutes of Health's National Institute of General Medical Sciences, which partly funded the research, "The process of transcription is essential to all living things so understanding how it initiates is enormously important. This work is a beautiful example of integrating multiple approaches to reveal the structure of a large molecular complex and provide insight into the molecular basis of a fundamental cellular process."

Nogales, who holds joint appointments with Berkeley Lab, the University of California (UC) at Berkeley, and the Howard Hughes Medical Institute (HHMI), is the corresponding author of a paper describing this study in the journal Nature. The paper is titled "Structural visualization of key steps in human transcription initiation." Co-authors are Yuan He, Jie Fang and Dylan Taatjes.

The fundamental process of life by which information in the genome of a living cell is used to generate biomolecules that carry out cellular activities is the so-called "central dogma of molecular biology." It states that genetic information flows from DNA to RNA to proteins. This straightforward flow of information is initiated by an elaborate system of proteins that operate in a highly choreographed fashion with machine-like precision. Understanding how this protein machinery works in the context of passing genetic information from DNA to RNA (transcription) is a must for identifying malfunctions that can turn cells cancerous or lead to a host of other problems.

Nogales and members of her research group used cryo-electron microscopy (cryo-EM), where protein samples are flash-frozen at liquid nitrogen temperatures to preserve their structure, to carry out in vitro studies of reconstituted and purified versions of the "transcription pre-initiation complex." This complex is a large assemblage of proteins comprised of RNA polymerase II (Pol II) plus a class of proteins known as general transcription factors that includes the TATA-binding protein (TBP), TFIIA, TFIIB, TFIIF, TFIIE and TFIIH. All of the components in this complex work together to ensure the accurate loading of DNA into Pol II at the start of a gene sequence.

"There's been a lack of structural information on how the transcription pre-initiation complex complex is assembled, but with cryo-EM and our in vitro reconstituted system we've been able to provide pseudo-atomic models at various stages of transcription initiation that illuminate critical molecular interactions during this step-by-step process," Nogales says.

The in vitro reconstituted transcription pre-initiation complex was developed by Yuan He, lead author on the Nature paper and a post-doctoral student in Nogales's research group.

"This reconstituted system provided a model for the sequential assembly pathway of transcription initiation and was essential for us to get the most biochemically homogenous samples," Nogales says. "Also essential was our ability to use automated data collection and processing so that we could generate all our structures in a robust manner."

Among the new details revealed in the step-by-step cryo-EM images was how the transcription factor protein TFIIF engages Pol II and promoter DNA to stabilize both a closed DNA pre-initiation complex and an open DNA-promoter complex, and also how it regulates the selection of a transcription start-site.

"Comparing the closed versus open DNA states led us to propose a model that describes how DNA is moved during the process of promoter opening," says He. "Our studies provide insight into how THIIH uses ATP hydrolysis as a source of energy to actually open and push the DNA to the active site of Pol II."

Nogales and her colleagues plan to further investigate the process of DNA loading into Pol II, as well as to include additional transcription factors into the assembly that are required for regulation of gene expression.

"Our goal is to actually build a structural model of the entire - more than two million daltons - protein machinery that recognizes and regulates all human DNA promoters," Nogales says. "For now we have the structural framework that's been needed to integrate biochemical and structural data into a unified mechanistic understanding of transcription initiation."
